Medicus Pharma reorients around new cancer drug candidate, sharpens focus for other programs

Medicus Pharma (NASDAQ:MDCX) is repositioning itself as a precision oncology-led biotechnology company, centering its strategy on CD228V, a second-generation antibody-drug conjugate targeting CD228-expressing solid tumors, the company said Tuesday.

CD228V is now the company's principal strategic priority, with initial development focused on establishing clinical proof-of-concept in a selected tumor population.

Medicus said melanoma, particularly in patients with relapsed or refractory disease, represents a compelling initial development opportunity based on the existing biological, translational and clinical foundation for the program.

The ADC targets melanotransferrin/CD228 in solid tumors and is supported by an active US Investigational New Drug application and a first-in-human Phase 1 clinical study along with prior scientific, translational and clinical development work.

Medicus said successful demonstration of clinical proof-of-concept in melanoma could lay the groundwork for further development in other CD228-expressing solid tumors.

The company is also sharpening its strategy for its other two therapeutic programs. For SkinJect, Medicus said it sees its strongest risk-adjusted opportunity in Gorlin syndrome rather than broad sporadic or nodular basal cell carcinoma lesions. The FDA has authorized the start of SKNJCT-005, the company's NDA-enabling registrational Phase 2b study for patients with Gorlin syndrome who present with multiple basal cell carcinomas.

Gorlin syndrome is a rare autosomal dominant disease in which patients can develop numerous basal cell carcinoma lesions over their lifetime, often requiring repeated surgical or other lesion-directed procedures. Medicus said concentrating SkinJect development on the condition could offer strategic advantages over the more competitive sporadic BCC market, including potential Orphan Drug Designation and Rare Pediatric Disease designation from the FDA, as well as favorable rare-disease-anchored net pricing.

For Teverelix, the company plans to prioritize its optimized, roughly 126-patient Phase 2 program in acute urinary retention along with PRECISION-E2, a Phase 2a study in women with symptomatic endometriosis. Medicus said it will continue advancing the scientific and strategic positioning of Teverelix in advanced prostate cancer through potential partnerships.

"Medicus has undergone a significant transformation this year," said Dr Raza Bokhari, CEO of Medicus.

Bokhari said the company is concentrating its capital and organizational resources on programs it believes offer the greatest potential for value creation and improved patient outcomes, while using strategic partnerships and other capital-efficient structures to preserve and unlock value in its other programs.

"We believe that our primary focus on precision oncology substantially enhances our long-term growth profile and positions the company to participate in one of the most dynamic sectors of biotechnology," he said.

Shares of Medicus added 5.1% on Tuesday morning.
