Mindset Pharma Inc (CSE:MSET, OTCQB:MSSTF, FRA:9DF) said that it has successfully demonstrated that its psilocybin analogs have a shorter duration and higher efficacy than typical psychedelics.
The Toronto-based company announced results from its preclinical studies on its Family 2 group of novel psilocybin/psilocin analogs and its Family 4 novel DMT/ 5-MeO-DMT analogs demonstrated superior efficacy, safety and shorter duration of action in head-to-head preclinical comparisons with their reference drugs.
Initial in vitro screening on the Family 2 compounds showed five- to 10-times the effect size as psilocin on human 5-HT2A receptors, which was verified through in vivo mouse head twitch assays, according to Mindset’s chief scientific officer Joseph Araujo.

Results also suggested that several of the compounds metabolized at least twice as rapidly as psilocin, Araujo added, while in vivo rodent studies suggested that the half-life of key Family 2 compounds were less than 50% of psilocybin.
For the Family 4 compounds, Araujo told investors that studies indicated greater effect size at the human 5-HT1A, 5-HT2A, and 5-HT2C subtype receptors compared to DMT and 5-MeO-DMT, according to Araujo. Results also suggested that the half-life of Mindset’s compounds range from seven to 90 minutes, providing a range of potential duration effects and differentiating the short-acting family from Mindset’s psilocybin-inspired families, Araujo added.
Mindset’s Family 2 compounds are next-generation psilocybin and psilocin analogs optimized for in-clinic settings, while Family 4 includes DMT and 5-MeO-DMT-inspired drug candidates that offer a broad range of pharmacological diversity suitable for in-clinic settings, the company said.
“The improvement in duration and efficacy/safety of our next generation compounds has meaningful implications for psychedelic-based therapy in the clinic,” Mindset CEO James Lanthier told investors.
“These improvements could potentially offer a scalable approach for psychiatrists as they manage dosing and treatment strategies for patients suffering from mental illness.”
