AtaiBeckley Inc. (NASDAQ:ATAI, XETRA:9VC) highlighted progress across its clinical pipeline and reaffirmed its financial outlook during its 2026 Virtual Investor Day, outlining plans to advance its lead candidate BPL-003 into Phase 3 trials for treatment-resistant depression while continuing development of additional mental health therapies.
The clinical-stage biotechnology company said it remains on track to initiate two parallel Phase 3 pivotal studies of BPL-003, a mebufotenin benzoate nasal spray, in the second quarter of 2026 following a successful End-of-Phase 2 meeting with the US Food and Drug Administration. The drug candidate has received Breakthrough Therapy Designation and is being developed as a rapid-acting treatment for patients with treatment-resistant depression.
The planned trials, named ReConnection-1 and ReConnection-2, will include a 12-week randomized, double-blind, placebo-controlled core study followed by a 52-week open-label extension that allows individualized retreatment based on pre-specified criteria.
“BPL-003 has demonstrated rapid, durable antidepressant effects, and the FDA’s supportive End-of-Phase 2 feedback enables us to advance into two parallel Phase 3 trials beginning in Q2 2026,” AtaiBeckley CEO Srinivas Rao said in a statement Tuesday.
“With Breakthrough Therapy Designation and a robust pivotal design, we believe BPL‑003 has the potential to meaningfully reshape the treatment landscape for TRD.”
AtaiBeckley said BPL-003 is supported by positive Phase 2b topline results in treatment-resistant depression, which showed rapid antidepressant effects as early as two days after a single dose and improvements that were maintained for up to eight weeks.
The company also reported that response and remission rates increased further following an optional second dose in the open-label extension phase.
Alongside clinical development, the company said it is advancing its commercial readiness strategy for BPL-003 with a treatment model designed to integrate into existing interventional psychiatry workflows, including those currently used for the depression therapy Spravato. According to AtaiBeckley, this approach is intended to allow clinics to incorporate the therapy without requiring in-session psychotherapy, with a focus on patient convenience and operational scalability.
“Our goal is to make BPL‑003 realistic and accessible in everyday clinical practice,” AtaiBeckley's senior vice president of product planning and early commercial development Kavita Panke said.
“People need treatments that work quickly, are durable, and don't require them to reorganize their lives. BPL-003’s short treatment window and intermittent dosing regimen could give clinics a practical and scalable way to reach far more patients.”
The company also reaffirmed that its current cash resources are expected to fund operations into early 2029, which would extend through the planned topline readouts from both Phase 3 studies.
Beyond BPL-003, AtaiBeckley highlighted continued progress across its broader pipeline. The company recently reported positive Phase 2a results for EMP-01 and expects topline Phase 2 data for VLS-01 in the second half of 2026.
The investor event concluded with a roundtable discussion featuring academic and clinical experts, including Dr. Peter Hendricks of the University of Alabama at Birmingham School of Public Health, Dr. David Feifel of the University of California, San Diego and the Kadima Neuropsychiatry Institute, and Dr. Samuel Wilkinson of Yale School of Medicine.
The discussion focused on the evolving treatment landscape for treatment-resistant depression and interventional psychiatry, including barriers to care, dosing approaches, and clinic infrastructure. The participants noted that shorter in-clinic treatment experiences and intermittent dosing models could help expand patient access while maintaining clinical effectiveness.
They also observed that the growing infrastructure supporting interventional psychiatry, driven in part by wider adoption of therapies such as Spravato, may help clinics accommodate emerging treatments like BPL-003.
The update sent AtaiBeckley shares around 5.5% higher on Tuesday morning in New York.
