Company Statement

 

Strategy

Xenetic’s strategy as a Speciality Drug Developer has at its core the Company’s three patented drug delivery platforms that allow the creation of novel and next generation drug and vaccine candidates with applications over a broad range of therapeutic areas.

 

While proprietary drug development in the orphan drug sector will be a first priority, the Company will continue to seek out-licensing arrangements with leading Big Bio and Big Pharma partners for the development of novel or next generation therapeutics.

 

Xenetic has already established a clinical record for potential blockbuster candidates for the treatment of anaemia and diabetes; and, through its various development agreements in Russia, India and elsewhere (such as that established in 2006 with Baxter Healthcare) the Company believes that the dissemination, application and commercialisation of its technologies will be better facilitated by the outlicensing of multiple product candidates currently beyond its economic capacity to develop in-house.

Xenetic has developed a number of proprietary patented technologies that are being used to create a new generation of drug and vaccine products with improved performance. Xenetic’s technologies include:

 

PolyXen® for protein drug delivery

PolyXen® is a versatile enabling technology that uses the biopolymer polysialic acid to prolong the active life and improve the pharmacokinetics and pharmacodynamics of therapeutic peptides and proteins, as well as conventional small molecule drugs.  PolyXen® is  a next generation method to PEGylation for the delivery of protein drugs.

 

ImuXen® for vaccine delivery

ImuXen® is an advanced enabling technology that uses liposome-based constructs to boost the effectiveness of DNA, protein and polysaccharide vaccines.  ImuXen® can achieve protective immunity in a single dose.

 

Oncohist

Oncohist™ has been established as a novel patent-protected platform technology that utilizes the special properties of human histone H1.3 as a critical component of the innate defense system for the treatment of cancer. An inducible expression system is used for large scale expression of Oncohist™ (recombinant human N-bis-met-Histone H1.3) in Escherichia coli.  Oncohist™ has strong anti-proliferative properties in cancer cells of different histological origins. This has been demonstrated extensively for hematologic malignancies, such as leukaemias, lymphomas and myelomas. Oncohist™ has also the potential application across a broad spectrum of other cancers.

 

PolyXen

PolyXen® is an enabling technology for protein drug delivery.  It uses the natural polymer polysialic acid (PSA) to prolong the active life and improve the stability of therapeutic peptides and proteins.  It can also be used for small molecule drugs.

Pre-clinical proof-of-concept studies and clinical studies in man have shown that the key benefits of PolyXen® for protein drug delivery include:

 

  • Preservation of drug functionality on conjugation
  • Improved stability in vivo
  • Prolonged pharmacological action
  • Prolonged active life of the drug in the blood circulation
  • Reduced frequency and amount of dosage
  • Reduced immunogenicity and antigenicity

ImuXen

ImuXen® is a liposomal technology platform designed to improve the delivery and effectiveness of DNA, protein and polysaccharide vaccines.

 

Proof of concept preclinical studies with a number of vaccines have shown that ImuXen® technology can help to generate the rapid onset of protective immune responses, in some cases following just a single dose.

 

The advantages of ImuXen® for DNA, protein and polysaccharide vaccines include:

 

 

  • Efficient delivery of vaccine antigens to the immune system
  • Enhanced immune responses
  • Protective immunity after a single injection
  • Protection of vaccine against degradation during storage

Oncohist

Oncohist™ exhibits the same properties as the body's own histone H1.3 molecules and has consequently low immunogenicity and antigenicity. Extensive data have shown that Oncohist molecules have the ability to destabilize selectively the tumor cell membrane which results in cell death. The selective attack of the membrane of tumor cells is due to the ability of Oncohist™ molecules to recognize the difference in membrane structure between tumor cells and their healthy counterparts. We believe that Oncohist™ has superior advantages over conventional cancer therapeutics, including high selectivity, low inherent toxicity, lack of induction of resistance and low immunogenicity.

 

A pilot clinical study (phase I/II) with Acute Myeloid Leukemia (AML) patients has been performed at the Saarland University Hospital at Homburg/Saar, Germany. The data showed that Oncohist™ is safe and well tolerated by patients while the first encouraging signs of clinical effect included three partial remissions, increase of thrombocytes and normalization of leukocytes as well as stabilization for several months in two patients.

 

Oncohist™ has been granted orphan drug status for Acute Myeloid Leukemia (AML) by both the FDA and EMA and additional orphan drug status for Acute Lymphocytic Leukemia (ALL) in Europe by the EMA.
